INS-1 is a rat insulinoma cell line that was originally isolated from pancreas tissue of the x-ray-induced insulinoma in a 2-year-old male rat. INS-1 exhibits the beta-cell-like morphology and adherent culture properties. These cells express a high level of insulin and are sensitive to glucose fluctuations. The INS-1 cells are heterogeneous, consisting of both mature insulin-positive cells and immature bi-hormonal cells that co-express insulin and glucagon. Therefore, INS-1 is a suitable cell line to investigate pancreatic islet beta-cell function and diabetes.

  The INS-1 cell line is an immortalized cells induced by radiation, isolated from pancreas of an x-ray-created transplantable insulinoma rats.

• What culture medium is recommended for INS-1 cells?

  The culture medium recommended for INS- 1 cells is composed of 1640 medium supplemented with 2.0 g/L Glucose, 2 mM L-Glutamine, 10 mM HEPES, 1 mM Sodium pyruvate, 1.5 g/L NaHCO3 to support optimal cell growth and maintenance, add 10% FBS.
  The sodium pyruvate, HEPES, and mercaptoethanol should be added at the specified concentration for cell growth.

• Why are my INS-1 cells do not attach?

  The INS-1 cell line show weak attachment at the beginning of the recovery, usually take 24-48 hours without changing medium. In the first few days, observing floating cells is normal. This cell also grow slowly, may form attached clumps. Please be patient and take care. Normally 3 days after thawing, the cells can be trypsinized and plated on 2 flasks to expand.
