SHP-77 expresses neuroendocrine markers and harbors KRAS and TP53 mutations,supporting studies of small cell lung cancer differentiation and immunotherapy.

Description

SHP-77 is a human small cell lung carcinoma (SCLC) cell line established in 1977 from a non-encapsulated primary tumor in the apical segment of the left upper lobe of the left lung of a 54-year-old Caucasian male patient. This cell line exhibits epithelial-like morphology with mixed growth properties, showing both loosely adherent clusters and suspension aggregates. Cytogenetic analysis reveals mutations in key oncogenic drivers including KRAS and TP53. SHP-77 expresses various neuroendocrine differentiation markers such as L-dopa decarboxylase, dense core secretory granules, neural cell adhesion molecule (NCAM/CD56), and surface antigens CD56/CD57 (HNK-1/Leu-7). The cell line demonstrates tumorigenic potential in immunocompromised murine models, making it valuable for studying SCLC progression and immunotherapy response.

Application

  • The SHP-77 cell line is a versatile research model widely used in immune-oncology and cytotoxicity studies related to small cell lung carcinoma (SCLC). It serves as an in vitro target in 51Cr and 111In release cytotoxicity assays, facilitating the study of cytotoxic immune responses. In vivo, SHP-77 is used in xenograft models to evaluate immune reactivity in immunodeficient mice, particularly using cells and serum from SCLC patients. Furthermore, they play a crucial role in assessing the immune status and therapeutic response in individuals with small cell lung carcinoma (SCLC) who have undergone radiation or chemotherapy treatments, offering valuable insights into treatment efficacy and resistance mechanisms.
