Description of TargetThis gene, a Kirsten ras oncogene homolog from the mammalian ras gene family, encodes a protein that is a member of the small GTPase superfamily. A single amino acid substitution is responsible for an activating mutation. The transforming protein that results is implicated in various malignancies, including lung adenocarcinoma, mucinous adenoma, ductal carcinoma of the pancreas and colorectal carcinoma. Alternative splicing leads to variants encoding two isoforms that differ in the C-terminal region.

  • 40 µg of whole cell lysate or 500 ng of in vitro expressed G12 WT and mutants were probed with KRAS G12D Mutant Recombinant Antibody [29B] (ATM00004) at 3 µg/ml.


  • KRAS G12D Mutant Recombinant Antibody [29B] (ATM00004) staining of PANC-1 (KRAS G12 Heterozygous), THP-1 (NRAS G12D Heterozygous) and HEK293 KRAS WT. Methanol fixed cells were probed with recAb at 1/50 dilution (20ug/ml) and visualized using anti Rabbit AF488



  • Surface-attached KRAS G12D Mutant Recombinant Antibody [29B] (ATM00004) at 50 µg/ml facilitated kinetic analysis via LSA Kinetic software, with two G12D antigen concentrations showing association and dissociation rates, ka = 2.7×10^3 M^-1s^-1 and kd = 1.4×10^-4 s^-1, respectively, and a 50 nM affinity constant (KD). No specific binding was observed for KRAS antigens with G12G or mutations G12S, G12C, G12R, and G12A



  • KRAS G12D Mutant Recombinant Antibody [29B] (ATM00004) rabbit Ab binding kinetics vs Human Leukocyte Antigen HLA-A11:01&B2M presenting KRAS- G12D peptide in 1x PBS buffer, pH 7.0. Serial dilutions of KRAS G12D Mutant Recombinant Antibody [29B] (ATM00004) rabbit antibodies from 50 ug/mL to 1.6ug/mL were used to attach the KRAS G12D Mutant Recombinant Antibody [29B] (ATM00004) to the surface for kinetic analysis via LSA Kinetic software fitting algorithm, with two G12D antigen concentrations showing association and dissociation rates, ka =2.78x10^3 M-1s-1 and kd = 5.83x10-5 s-1, respectively, and a 23.4 nM affinity constant (KD). Sequential testing with 50 nM and 100 nM of G12D antigens yielded these kinetic parameters, represented by blue and green curves. No specific binding was observed for KRAS antigens with G12G or mutations G12S, G12C, G12R, and G12A
