Human Orbital Fibroblasts (HOFs) are primary cells isolated from human orbital connective tissue and cryopreserved following primary culture, and are commonly referenced in orbital and autoimmune research as representative primary fibroblasts, particularly in studies requiring non-transformed fibroblast models from ocular tissues. These fibroblasts exhibit a typical spindle-shaped morphology and adhere well in vitro. HOFs express markers such as vimentin, CD90, and TSH receptor (TSHR), and exhibit phenotypic plasticity that allows differentiation into adipocytes or myofibroblasts. They play an essential role in maintaining the structural integrity of orbital tissue and are actively involved in extracellular matrix remodeling. Research has shown their significant involvement in Graves’ ophthalmopathy (GO), where they respond to inflammatory stimuli by producing cytokines such as IL-6 and interact with endothelin receptors, contributing to disease pathogenesis. These characteristics make them valuable for studying orbital tissue biology and related autoimmune disorders. Repeated freezing and thawing should be avoided during culture to maintain cell viability and function.
