Introduction

The Mitochondria Activity Assay kit utilizes tetramethylrhodamine ethyl ester (TMRE), a cell permeable red-orange dye to measure differences in mitochondria membrane potential. Since TMRE is positively charged, it accumulates in active mitochondria, which has a negative charge. In contrast, inactive mitochondria will have less TMRE binding and a lower signal due to its decreased membrane potential.

Materials provided with the kit

• 8x12 96-well plate

• 1mM TMRE (-20oC)

• 20mM FCCP (-20oC)

• KRPG buffer (RT)

Material required but not provided

• Fluorescence plate reader capable of measuring fluorescence at 575 nM

Reagent preparation before starting experiment

• Dilute the 1mM TMRE 1:1000 in cell media to make 1uM TMRE

• Dilute the 20mM FCCP 1:1000 in cell media to make 20uM FCCP

Assay procedure

1. Grow adherent cells the day before the experiment.

2. Harvest the cells and plate them on a clear 96-well plate and allow them to adhere overnight.

3. Treat the cells using desired compounds/conditions. Use FCCP as a negative control. Incubation times will vary depending on treatment. (1 hr for FCCP)

4. Remove cell media and wash with PBS.

5. Stain cells with cell media containing TMRE. Incubate for 30 minutes.

6. Remove cell media and wash with PBS.

7. Add KPRG buffer to each well.

8. Measure the plate in a fluorescence plate reader at an excitation wavelength and emission wavelength of 549nM and 575nM, respectively
