Novel insight on Thyroid Stimulating Hormone Receptor

Thyroid hormone is one of the most important metabolic hormones. When the hypothalamus is stimulated, thyroid hormone-releasing hormone (TRH) is released, and the secretion of TRH further stimulates the pituitary to release thyroid stimulating hormone (TSH). Thyroid stimulating hormone receptor (TSHR) is a receptor for TSH, activated TSHR can be coupled to the stimulatory Gs protein to stimulate the production of thyroid hormone. TSHR a subunit can be dissociated from the B subunit to become an antigen and could become an autoantigen that induces various types of antibodies against TSHR. Autoantibodies (such as M22 or K1-70) can cause abnormal secretion of TSHR and lead to diseases such as Graves’ disease or hypothyroidism. These diseases are affecting hundreds of millions of patients worldwide, however, the basis of TSH and antibody-mediated TSHR activation is unclear. Owing to its disease relevance, TSHR is an attractive target for drug discovery.

Flow-cytometry
Flow-cytometry using anti-human TSHR antibody.TSHR Transfected CHO cells were stained with an irrelevant antibody (Blue Histogram) or an anti-human TSHR antibody monoclonal antibody (Catalog # FHD18140 ,Green Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a FITC conjugated goat anti-mouse antibody (Catalog # PMB96441) and cells analysed on a NovoCyte Flow Cytometer.
Western Blot
Various lysates were subjected to SDS PAGE followed by western blot with TSHR antibody (FHD18140) at 1 µg/ml.

Lane 1: Mouse ovary lysate

Second Ab: Goat Anti-Mouse IgG H&L Polyclonal antibody, HRP (PMB96431) at 0.1 μg/mL.
