About Elite™ Extracellular Nitric Oxide (NO) Assay Kit

Nitric oxide (NO) is a short-lived gaseous signaling molecule that regulates numerous physiological processes, including vascular homeostasis, neurotransmission, immune responses, inflammation, and cell-to-cell communication. It is synthesized from L-arginine by the nitric oxide synthase (NOS) family of enzymes, including endothelial NOS (eNOS)neuronal NOS (nNOS), and inducible NOS (iNOS). Because nitric oxide rapidly diffuses across cell membranes and has an extremely short half-life, extracellular NO levels provide a valuable indicator of cellular nitric oxide production and biological activity. Dysregulated NO production has been associated with cardiovascular disease, chronic inflammation, neurodegenerative disorders, cancer, infection, and autoimmune diseases, making accurate measurement of extracellular nitric oxide an important tool in biomedical research.

The Elite™ Extracellular Nitric Oxide (NO) Assay Kit provides a rapid, sensitive, and homogeneous assay for the quantitative detection of extracellular nitric oxide released from cultured cells. The assay employs a highly specific nitric oxide detection chemistry that reacts with extracellular NO to generate a stable, measurable signal while minimizing interference from other reactive oxygen species (ROS). Its simple add-and-read workflow requires no wash steps and is readily compatible with 96- and 384-well microplates, enabling both routine laboratory measurements and high-throughput screening applications. The assay is well suited for monitoring nitric oxide production from endothelial cells, macrophages, neurons, and other NO-producing cell types following stimulation with cytokines, inflammatory mediators, or candidate therapeutic compounds.

Biological Significance of Extracellular Nitric Oxide

Nitric oxide functions as a versatile signaling molecule that regulates vascular tone, platelet aggregation, neurotransmission, host defense, and immune cell activation. In endothelial cells, eNOS-derived NO promotes vasodilation and maintains cardiovascular homeostasis. During inflammation, activation of iNOS in macrophages and other immune cells results in sustained NO production, contributing to antimicrobial defense as well as inflammatory tissue injury when produced in excess. Because nitric oxide readily diffuses into the extracellular environment before being converted into more stable oxidation products, extracellular NO measurements provide a direct and physiologically relevant assessment of cellular nitric oxide production.
