About Elite™ Rapid Glutathione (GSH/GSSG) Ratio Assay Kit

Glutathione is the most abundant intracellular antioxidant and plays a critical role in maintaining cellular redox homeostasis. It exists primarily in the reduced form (GSH) under normal physiological conditions, where it protects cells by scavenging reactive oxygen species (ROS), detoxifying xenobiotics, and maintaining protein thiol groups in their reduced state. During oxidative stress, GSH is oxidized to glutathione disulfide (GSSG) by glutathione peroxidases. Consequently, the GSH/GSSG ratio is one of the most widely accepted indicators of cellular oxidative stress and redox balance. A decrease in this ratio reflects increased oxidative damage and has been associated with aging, cancer, neurodegenerative diseases, cardiovascular disorders, diabetes, inflammation, and drug-induced toxicity.

The Elite™ Rapid Glutathione (GSH/GSSG) Ratio Assay Kit provides a rapid, ultrasensitive fluorometric method for quantifying reduced glutathione (GSH)total glutathione (GSH + GSSG), and calculating the GSH/GSSG ratio in biological samples. The assay utilizes a proprietary non-fluorescent probe that becomes highly fluorescent upon reacting with GSH, enabling detection of as little as 1 picomole of glutathione in a 100 μL assay volume. The simple one-step, mix-and-read protocol requires minimal sample handling and is readily adaptable to 96- and 384-well microplate formats without separation steps. Fluorescence is measured at Ex/Em = 490/520 nm, making the assay ideal for routine laboratory studies as well as high-throughput screening (HTS). Suitable sample types include cell lysates, tissue homogenates, serum, plasma, urine, and other biological samples.

Biological Significance of the GSH/GSSG Ratio

Glutathione serves as the central component of the intracellular antioxidant defense system. Under physiological conditions, more than 90% of cellular glutathione exists in the reduced (GSH) form, while only a small fraction is present as oxidized glutathione (GSSG). During oxidative stress, reactive oxygen species consume GSH through glutathione peroxidase-mediated detoxification reactions, resulting in the accumulation of GSSG. The GSH/GSSG ratio therefore provides a highly sensitive and physiologically relevant indicator of cellular redox status. A reduced ratio reflects oxidative stress, mitochondrial dysfunction, impaired antioxidant capacity, and activation of cell death pathways, including apoptosis and ferroptosis. Alterations in the GSH/GSSG ratio have been implicated in numerous pathological conditions, including cancer, Alzheimer's disease, Parkinson's disease, cardiovascular disease, diabetes, liver injury, chronic inflammation, and aging.
