About Elite™ Lipid Peroxidation Assay Kit

Lipid peroxidation is a hallmark of oxidative stress that occurs when reactive oxygen species (ROS) attack polyunsaturated fatty acids within cellular membranes, leading to the formation of lipid hydroperoxides and reactive aldehydes. Excessive lipid peroxidation disrupts membrane integrity, alters membrane fluidity, impairs cellular signaling, and ultimately contributes to cell dysfunction and death. This process plays a central role in numerous pathological conditions, including ferroptosis, neurodegenerative diseases, cardiovascular disorders, diabetes, inflammation, ischemia-reperfusion injury, aging, and cancer. Consequently, sensitive detection of lipid peroxidation has become an essential tool for investigating oxidative damage, evaluating antioxidant activity, and studying redox biology.

The Elite™ Lipid Peroxidation Assay Kit provides a rapid, sensitive, and homogeneous fluorescent assay for detecting lipid peroxidation in live cells. The assay employs a lipid peroxidation-sensitive fluorescent probe that readily incorporates into cellular membranes. Upon oxidation by lipid peroxides, the probe undergoes a fluorescence shift, enabling quantitative measurement of lipid peroxidation in living cells. The assay features a simple mix-and-read protocol without extraction or complex sample preparation and is compatible with fluorescence microplate readers, fluorescence microscopy, and flow cytometry. Optimized for 96- and 384-well microplates, the kit provides an excellent platform for routine oxidative stress studies as well as high-throughput screening (HTS) of compounds that induce or inhibit lipid peroxidation.

Applications

Biological Significance of Lipid Peroxidation

Lipid peroxidation is initiated when reactive oxygen species oxidize membrane phospholipids containing polyunsaturated fatty acids, generating lipid hydroperoxides and secondary reactive products that amplify oxidative damage throughout the cell. Under physiological conditions, antioxidant defense systems—including glutathione (GSH), glutathione peroxidase 4 (GPX4), vitamin E, and other lipid-soluble antioxidants—maintain membrane integrity by limiting lipid peroxide accumulation. When these protective mechanisms are overwhelmed, excessive lipid peroxidation promotes membrane disruption, mitochondrial dysfunction, inflammatory signaling, and regulated cell death pathways, particularly ferroptosis, an iron-dependent form of programmed cell death driven by lipid peroxide accumulation. Lipid peroxidation has therefore emerged as a critical biomarker for studying oxidative injury and therapeutic responses across a broad range of diseases.
